  • No fetal adverse effects for amiloride in animals Limited human data but reported association with hypospadias in 1st trimester exposure and with mild intrauterine growth retardation if used throughout pregnancy.
  • No adequate human data for hydrochlorothiazide, no teratogenic effect has been reported but in general thiazides can cause fetal bradycardia and fetal death by causing maternal hypotension, neonatal hypoglycemia, hyponatremia, hypotonia and thrombocytopenia.
  • Thiazides also inhibit labor by affecting uterine smooth muscles.

  • Studies in rats have shown that amiloride is excreted in milk in concentrations higher than those found in blood, but it is not known whether amiloride HCl is excreted in human milk.
  • However, thiazides appear in breast milk. Because of the potential for serious adverse reactions in nursing infants, a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.
