  • Dapsone is carcinogenic in animals.
  • Causes blood dyscrasias in adults (mainly hemolytic anemia).
  • No well - controlled data in human pregnancy. Reported to cause hemolytic anemia in the mothers and their newborns.
  • Reported single case of cleft palate is a split in roof of the mouth, due to failure of two sides to fuse during embryonic development. Could also be a cleft lip, which is a split in the lip">cleft palate and single case of congenital hip dislocation in two infants exposed to sulfone drugs (including dapsone) in pregnancy can cause neonatal hyperbilirubinemia.
  • Pyrimethamine, is teratogenic in animals causing cleft palate is a split in roof of the mouth, due to failure of two sides to fuse during embryonic development. Could also be a cleft lip, which is a split in the lip">cleft palate limb defects, neural tube defects and chromosomal abberations.
  • It is Folic acid antagonist, should be avoided in the 1st trimester and Folic acid supplementation should be given during therapy.
